IGF-1 DES, or des(1-3)IGF-1, is human insulin-like growth factor 1 with its first three residues removed, a 67-residue protein held together by three disulfides. This listing is a preloaded autoinjector pen containing 3 mg of it in 3 mL of bacteriostatic water. Studies of the truncated protein have dealt with its potency in cultured muscle cells, its weak binding to IGF-binding proteins and its fate in rat plasma.
What is in the pen
The 3 mL cartridge holds sterile solution with IGF-1 DES at 0.1% w/v: 1 mg/mL and 3 mg per pen, or roughly 0.14 mM at the folded mass of 7365.4 Da. It is dissolved in bacteriostatic water preserved with 0.9% benzyl alcohol. On the safety data sheet the protein is not classified, while benzyl alcohol carries warnings for harm if swallowed or inhaled, eye irritation and skin sensitisation. Store the pen refrigerated at 2 to 8 °C in its carton and away from light, keep it capped once started, and do not let it freeze; a frozen, discoloured or cloudy solution should be discarded. The IGF-DES 2 mg vial supplies the same protein freeze-dried, and peptide pen vs vial sets out the general differences.
What the research covers
The truncated form was first isolated from bovine colostrum, where it occurred alongside IGF-1 and IGF-2 and differed from IGF-1 only by lacking the N-terminal Gly-Pro-Glu1. In cultured L6 myoblasts, destripeptide IGF-1 stimulated protein synthesis at roughly seven-fold the potency of IGF-1, whether purified or chemically synthesised, but competed no better for IGF-1 binding to the cells2.
The explanation lies with the IGF-binding proteins. Ballard and colleagues found that in rat plasma the labelled truncated protein barely associated with binding proteins, stayed mostly free in the circulation and was cleared far faster than IGF-I or IGF-II, with a larger volume of distribution3. More of the radioactivity reached the adrenals, brain, skin and gut with des(1-3)IGF-I than with the other two peptides3.
Frequently asked questions
How does the pen compare with the 2 mg vial?
The vial supplies 2 mg of dry protein that stays at −20 °C until a laboratory dissolves it. The pen arrives as a solution, 3 mg in 3 mL at 1 mg/mL, and is kept refrigerated. Both contain the same 67-residue des(1-3)IGF-1.
Is IGF-1 DES the same as IGF-1 LR3?
No. DES removes the first three residues of IGF-1, while LR3 adds a 13-residue extension and changes residue 3. They have different sequences and masses and are sold as separate products.
