Kisspeptin-10 (KP-10) is the ten-residue C-terminal fragment of human kisspeptin, YNWNSFGLRF-NH2, and an agonist at the kisspeptin receptor. In this listing it comes as a sterile solution in bacteriostatic water, 10 mg in a metered spray bottle. Kisspeptin research covers the receptor, its control of GnRH and gonadotropin release, and human studies that used other routes; none of the work cited here tested a nasal spray.

What is in the bottle

The bottle contains kisspeptin-10 dissolved in bacteriostatic water, with 0.9% benzyl alcohol as the preservative. The safety data sheet gives 10 mg of peptide per bottle at no more than 0.1% w/v, and the bottle label states the concentration. The peptide is the same H-YNWNSFGLRF-NH2 as in the kisspeptin-10 5 mg vial, with a theoretical mass of 1302.4 Da. Kisspeptin-10 carries no hazard classification on the SDS; benzyl alcohol is listed as harmful if swallowed or inhaled, a serious eye irritant and a skin sensitiser.

What the research covers

Kisspeptin was first isolated from human placenta as metastin, a 54-residue peptide with an amidated C-terminus, encoded by the KiSS-1 metastasis suppressor gene and recognised by an orphan receptor then called hOT7T1751. In that 2001 paper metastin inhibited chemotaxis and invasion of receptor-transfected CHO cells and reduced lung metastasis of receptor-transfected B16-BL6 melanoma cells in vivo1. Kisspeptin-10 is the last ten residues of that peptide.

The short fragment does not last long in circulation. In male mice its half-life in the bloodstream was about 4 minutes, against about 32 minutes for kisspeptin-54, and it sustained LH release for a much shorter time2. In men, kisspeptin-54 had a plasma half-life of 27.6 minutes in a small double-blind, placebo-controlled crossover study with intravenous infusion that measured LH, FSH and testosterone3.

No paper cited here delivered kisspeptin-10 through the nose, so how much crosses the nasal lining, and whether it survives long enough to reach its receptor, is not established. General evidence on the route is in the overview of peptide nasal sprays, and the receptor and cell work is in the kisspeptin-10 (kisspeptin 45-54) guide.

Frequently asked questions

Is the spray the same peptide as the kisspeptin-10 vials?

Yes. All of them hold kisspeptin-10, H-YNWNSFGLRF-NH2, CAS 374675-21-5. The vials contain freeze-dried powder stored at −20 °C; the spray contains 10 mg already dissolved in bacteriostatic water and is stored at 2 to 8 °C, never frozen.

Has kisspeptin-10 been studied as a nasal spray?

Not in any paper cited on this page. The human studies here used intravenous and other non-nasal routes, and the mouse data show kisspeptin-10 clears from the blood within minutes. There are no published findings for this format to point to.
