Kisspeptin-10, the amidated ten-residue fragment of kisspeptin (YNWNSFGLRF-NH2), activates the kisspeptin receptor that drives GnRH release. This preloaded pen carries 10 mg of kisspeptin-10 in 3 mL of bacteriostatic water, about 3.33 mg/mL, so there is no powder to make up. Research on the peptide includes receptor and reproductive physiology, but for a solution product the most relevant papers are the ones on how quickly kisspeptin-10 breaks down.

What is in the pen

Each pen holds a 3 mL glass cartridge of sterile solution. The safety data sheet lists kisspeptin-10 at 0.33% w/v, 10 mg per pen, in bacteriostatic water with 0.9% benzyl alcohol as an antimicrobial preservative. The peptide is the same H-YNWNSFGLRF-NH2 sold freeze-dried in the kisspeptin-10 5 mg vial, theoretical mass 1302.4 Da, so the pen holds about 7.7 µmol, close to 2.6 mM. The SDS does not classify the peptide. Benzyl alcohol is listed as harmful if swallowed or inhaled, a serious eye irritant and a possible skin sensitiser.

Store the pen at 2 to 8 °C in its carton, away from light, and keep it capped once opened. Do not freeze it; discard a pen whose solution has frozen, changed colour or turned cloudy. How pens and vials compare in general is covered in peptide pen vs vial.

What the research covers

Kisspeptin-10 is the smallest kisspeptin fragment that keeps activity at the kisspeptin receptor, which signals through Gq/11 and phospholipase C1. Its stability is the main question for a ready-made solution. Liu and colleagues developed an LC-MS/MS assay for kisspeptin-10 in rat plasma and, in stability studies run with it, found that the peptide decomposed with half-lives of 6.8 minutes at 4 °C, 2.9 minutes at 25 °C and 1.7 minutes at 37 °C2. The principal breakdown product was kisspeptin-10 without its N-terminal tyrosine2. The abstract does not spell out the medium for those stability runs, but the assay was built for rat plasma, and nothing in it describes sterile bacteriostatic water.

The sequence also carries two asparagines. Asparagine deamidation in peptides has been measured directly by mass spectrometry: Robinson and Robinson determined rates for 306 asparaginyl sequences in model peptides at pH 7.4 and 37 °C, and used the method to measure how protein structure affects that rate3. That work gives no figure for kisspeptin-10 itself, and no study in these references followed kisspeptin-10 in a preserved aqueous solution, so the shelf life of the pen solution is not established here. The safety data sheet's storage conditions are the ones to keep to.

The receptor biology, the volunteer studies and the cell work are summarised in the kisspeptin-10 (kisspeptin 45-54) guide.

Frequently asked questions

How does the pen differ from the kisspeptin-10 vials?

The 5 mg and 10 mg vials hold freeze-dried kisspeptin-10 kept at −20 °C until a laboratory dissolves it. The pen holds 10 mg already in 3 mL of bacteriostatic water, about 3.33 mg/mL, and is kept at 2 to 8 °C. The peptide and its sequence are the same in all three.

How long is kisspeptin-10 stable in the pen?

No study cited here has measured it in bacteriostatic water. A 2013 study found the peptide broke down within minutes under the conditions of its rat plasma assay, losing the N-terminal tyrosine first. Keep the pen at 2 to 8 °C, dark and unfrozen, as the safety data sheet requires.
