This PT-141 nasal spray holds bremelanotide, the cyclic melanocortin peptide first developed under the code PT-141, as a ready-made solution. Each metered spray bottle contains 10 mg of bremelanotide in bacteriostatic water, so there is no powder to dissolve. Bremelanotide research concerns its design as an alpha-MSH analogue, its action at melanocortin receptors in the brain and controlled trials that used a different route, which leaves the nasal format with little published data of its own.

What is in the bottle

The bottle holds a sterile aqueous solution of bremelanotide in bacteriostatic water, with 0.9% benzyl alcohol as an antimicrobial preservative. The safety data sheet gives the peptide as 10 mg per bottle at no more than 0.1% w/v. It is the same compound as the PT-141 10 mg vial: Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, theoretical mass 1025.2 Da. The SDS does not classify the peptide as hazardous. Benzyl alcohol carries statements for harm if swallowed or inhaled, serious eye irritation and possible allergic skin reactions.

Keep the bottle upright and capped at 2 to 8 °C, away from light, and do not let it freeze. General guidance on peptides in solution is in peptide storage and stability.

What the research covers

Bremelanotide is a synthetic analogue of alpha-MSH and an agonist at melanocortin receptors, among them MC3R and MC4R, which are expressed mainly in the central nervous system1. In rats, systemic PT-141 raised c-Fos immunoreactivity in hypothalamic neurons, evidence that it reached and activated cells there1. Palatin Technologies developed it as an analogue of melanotan II, itself a truncated cyclic alpha-MSH analogue2.

The main human trials did not use a nasal spray. RECONNECT, two identical 24-week phase 3 trials in premenopausal women with hypoactive sexual desire disorder, compared a non-nasal bremelanotide formulation with placebo and measured desire and distress scores as co-primary endpoints3.

Frequently asked questions

Is PT-141 nasal spray the same peptide as the PT-141 vial?

Yes. Both contain bremelanotide, the cyclic heptapeptide with CAS number 189691-06-3. The vial holds 10 mg as freeze-dried powder kept at −20 °C. The spray holds 10 mg already dissolved in bacteriostatic water and is kept cold at 2 to 8 °C, never frozen.

How much bremelanotide does the bottle contain?

10 mg per bottle, recorded on the safety data sheet at no more than 0.1% w/v, with 0.9% benzyl alcohol as the preservative. The label on the bottle states the amount and concentration for the bottle in hand.

Has bremelanotide been studied as a nasal spray?
